Analytical flow cytometers

The Flow Cytometry Facility is eqquiped with conventional and full spectrum flow cyometers.

2x Aurora Full Spectrum Flow Cytometer (Cytek biosciences) 

Our facility is equipped with two 5-laser instruments. One of whch is capable of sampling form a temperature controlled 96-well plate

The Aurora is a 5-laser Full Spectrum Flow Cytometer that can be used to explore the wide range of immune cell types that are involved in combatting cancer. Compared to conventional flow cytometers, this instrument allows for deeper probing of cell function and make it easy to investigate cells that show relatively high auto-fluorescence.

BD FACSSymphony A5 SORP (BD Biosciences)

The FACSSymphony A5 is a custom ordered 32 parameter flow cytometer that is used to explore the wide range of immune cell types that are involved in combatting cancer, and their functions.
It allows for simultaneous detection of 30 fluorochromes and with low electronic noise, it can detect very dim signals.

This feature also makes it ideal for the application of the combinatorial coding method to investigate specific CD8 T cell responses to a large number of cancer associated neo-antigens simultaneously.

BD LSR-II SORP (BD Biosciences)

Like the FACSSymphony A5 SORP, the LSR-II SORP is a custom ordered flow cytometer that is equipped with 5 lasers and can detect up to 18 parameters simultaneously.

2x BD LSRFortessa (BD biosciences)

We have 2 Fortessa flow cytometers. 

Both flow cytometers are equipped with 4 lasers, and can detect up to 16 or 18 fluorescent labels simultaneously. One of which is capable of sampling fomr 96-well and 384-well plates.

These instruments are the work horses of our Facility. As such, their optical configurations ensure a large degree of flexibility in fluorophore choice.

Attune NxT Acoustic Focusing Cytometer (Thermo Fisher Scientific)

The Attune NxT is a 3-laser system that allows the use of up to fluorochromes simultaneously. The instrument is designed to maintain resolution at high flow rates. This makes the Attune NxT excellently suited for rare event detection. Additionally, the instrument is used for cell cycle analysis and absolute cell counts from whole blood samples.

Intellicyt® iQue Screener PLUS (Sartorius)

The iQue Screener PLUS is excellently suited to perform screens from a large number of samples with small sample volumes. One can think of small molecule or drug discovery assays. The instrument is also excellent for bead based assays.

The instrument is currently located in the Robotics & Screening center. 

The iQue has 3 lasers and can detect up to 13 different fluorophores simultaneously.

Cell sorters

The Flow Cytometry Facility is eqquiped with a total of 5 cell sorters: 

  • FACSDiscover S8 (BD biosciences)
  • Aurora CS cell sorter (Cytek biosciences)
  • 3 FACSAria TM Fusions  (BD biosciences)

All sorters are located inside biological safety cabinets. This allows us to perform cell sorting under BSL II+ (ML-II+) biosafety conditions.

FACSDiscover S8 cell sorter (BD biosciences)

The BD FACSDiscover™ S8 Cell Sorter is a spectral flow cytometry based sorter with five lasers and 78 detection channels, enabling full fluorescence spectrum measurement.

It is the first system to combine spectral flow sorting with real-time image-based analysis. Using BD CellView™ Image Technology and BD SpectralFX™ Technology, it integrates spectral, spatial, and morphological data to enhance cell identification and sorting.

Aurora CS (Cytek biosciences)

The detection method is based on Full Spectrum Profiling. This enables us to discriminate fluorophores that are very simlar, which allows for panels up to 50 parameters. More importantly, the system allows for flexibility in the panel design.

The sorter can be used for 6-way sorting in tubes and sorting into 96-well plates . 

This cell sorter is equipped with 5 lasers:

  • 355nm; 405nm; 488nm; 561nm; 640nm

3 FACSAria Fusions (BD Biosciences)

The FACSAria Fusions are cuvette-based sorters that are equipped with 3, 4 or 6 lasers, and can detect from 9 to 18 different fluorescent labels simultaneously. The sorters can be set-up to sort cells up to 25-30 micrometer in diameter when in single cell suspension. Four cell population subsets may be collected simultaneously from a single sample when sorting into Eppendorf of 5ml tubes for bulk sorting. Alternatively, single cells or small number of cells can be sorted into plates, from 6-well to 384 well plates.
